The Response of Leaf Explants Growth of Tobacco (Nicotiana tabacum L.) PRANCAK-95 in Various Concentration of Sucrose and BAP In Vitro

Popis: This research aims to determine the response of sucrose and BAP on tobacco (Nocotiana tabacum L.) Prancak-95 growth in vitro. This research was using Randomized Complete Block Design (RCBD) with 2 factors and 3 replications. the first factor was sucrose consisted 3 levels, there were 20 gr/l, 30 gr/l and 40 gr/l. The second factor was BAP consisted 3 levels, there were 2 ppm, 3 ppm and 4 ppm. It was conducted at Tissu Culture Laboratory of State Polytechnic of Jember in August to December 2018. The data were analyzed by using Analysis of Variance (ANOVA) and followed by Duncan’s Multiple Range Test (DMRT) 5%. The parameters were early callus, fresh weight of callus, texture of callus, early budding, fresh weight of shoot, height of shoot and number of shoot. The results of this research show that application of sucrose 20 gr/L is significant effect on height of shoot. The application of BAP 4 ppm is significant effect on early budding, height of shoot and fress weight of callus. Concentration of 3 ppm BAP is significant effect on fresh weight of shoot and number of shoot. The interaction between sucrose 20 gr/L and BAP 4 ppm is significant effect on fresh weight of shoot. Also the interaction between sucrose 30 gr/L and BAP 3 ppm is significant effect on number of shoot.
Penelitian ini ditujukan untuk mengetahui kombinasi antara sukrosa dan BAP, terhadap pertumbuhan tembakau (Nicotiana tabacum L.) Prancak-95 secara in vitro. Penelitian ini menggunakan Rancangan Acak Kelompok (RAK) Faktorial yang tersusun atas 2 faktor dan 3 ulangan. Faktor I adalah Sukrosa yang terdiri dari 3 taraf yaitu 20 gr/l, 30 gr/l dan 40 gr/l. Faktor II adalah BAP yang terdiri dari 3 taraf yaitu 2 ppm, 3 ppm dan 4 ppm. Penelitian tersebut dilakukan di Laboratorium Kultur Jaringan Politeknik Negeri Jember pada bulan Agustus 2018 - Desember 2018. Data yang didapat kemudian dianalisis memakai Analysis Of Variance (ANOVA) dan dilteruskan dengan DMRT taraf 5%. Parameter pengamatan meliputi kedinian kalus, berat basah kalus, tekstur kalus, kedinian tunas, berat basah tunas, tinggi tunas dan jumlah tunas. Hasil penelitian menunjukkan bahwa Pengaruh konsentrasi sukrosa 20 gram/liter berbeda nyata tehadap parameter tinggi tunas. Pengaruh konsentrasi BAP 4 ppm berbeda nyata terhadap parameter kedinian tunas, tinggi tunas dan berat basah kalus. Selain itu pengaruh konsentrasi BAP 3 ppm berbeda nyata terhadap parameter berat basah tunas, dan jumlah tunas. Pengaruh konsentrasi sukrosa 20 gram/liter dan BAP 4 ppm berbeda nyata terhadap parameter berat basah tunas. Selain itu pengaruh konsentrasi sukrosa 30 gram/liter dan BAP 3 ppm berbeda nyata terhadap parameter jumlah tunas.
