Serum dependence of expression of the transformed phenotype: Experiments with subline of mouse L fibroblasts adapted to growth in serum-free medium

Popis: | Characteristics of LSF cells grown in serum-containing and serum-free medium were compared. LSF is a subline of the L-strain of mouse transformed fibroblasts adapted to continuous growth in serum-free medium. Proliferation of LSF cells in monolayer on solid substratum was almost similar in serum-containing and in serum-free media. However, several other characters were found to be altered by the addition of serum to the serum-free medium: ability of cells to form colonies in semi-solid medium increased considerably; agglutinability of cells by Concanavalin A increased; uptake of 2-deoxy-D-glucose by the cells increased considerably; ability of cells to metabolize benzo (a)pyrene was inhibited; cell morphology was altered and, in particular, the cells became less spread on the substratum and density of microvilli on the cell surface increased. All these changes induced by serum were reversed by transfer of the cells back into serum-free medium. Thus, addition of serum increased the expression of a number of cellular traits characteristic of transformed phenotype, while in serum-free medium partial phenotypic reversion of transformation was observed. A possible role of serum in the expression of the transformed phenotype is discussed. It is pointed out that cell lines adapted to growth in serum-free medium provide an experimental system convenient for analysis of the effects of different serum components on the cell phenotype. Serodependance De L'expression Du Phenotype Transforme: Experiences Avec Une Sous-lignee De Fibroblastes Murins L Adaptee A La Croissance En Milieu Sans Serum Les auteurs ont compare les caracteristiques des cellules LSF cultivees dans un milieu contenant du serum et dans un milieu n'en contenant pas. Les LSF constituent une sous-lignee de la souche L de fibroblastes murins transformes adaptee a la croissance continue en milieu sans serum. La proliferation des cellules LSF en monocouches sur substrat solide est assez semblable dans les deux types de milieux. Toutefois, on a constate que plusieurs autres caracteristiques sont alterees par l'addition de serum au milieu qui n'en contient pas: l'aptitude des cellules a former des colonies en milieu semi-solide a considerablement augmente; leur agglutinabilite par la Concanavaline A s'est accrue; l'incorporation de 2-desoxy-D-glucose s'est sensiblement intensifiee; leur aptitude a metaboliser le benzo(a)pyrene a ete inhibee; leur morphologie a ete alteree et, en particulier, les cellules se sont moins disseminees sur le substrat et la densite des microvillosites de la surface s'est accrue. On observe une reversion de toutes ces alterations induites par le serum lorsque les cellules sont replacees dans un milieu sans serum. L'addition de serum a donc intensifie l'expression d'un certain nombre de particularites cellulaires caracteristiques du phenotype transforme tandis qu'en milieu sans serum, on a observe une reversion phenotypique partielle de la transformation. Les auteurs examinent le rǒle possible du serum dans l'expression du phenotype transforme. Ils soulignent que les lignees cellulaires adaptees a la croissance en milieu sans serum constituent un systeme experimental qui se prěte bien a l'analyse des effets des differents composants du serum sur le phenotype cellulaire. |
