Oncogenic transformation of chick-embryo fibroblasts by Rous sarcoma virus alters rubidium uptake and ouabain binding

Popis: | The ouabain-sensitive movement of monovalent cations has been determined in chick embryo fibroblasts and in cells infected by viruses such as Schmidt-Ruppin strain of sub-group A (SR-RSV), a temperature-sensitive mutant of this virus or Rous associated virus of sub-group A (RAV). The velocity of the ouabain-sensitive cation movement was estimated by measuring 86Rb+ uptake and the number of cation translocating macromolecules were determined by measuring [3H]ouabain binding to untransformed and oncogenically transformed chick embryo fibroblasts. There was a parallel decrease in 86Rb+ uptake and [3H]ouabain binding as a result of cell oncogenic transformation. There was a reciprocal relationship between the decrease in these two parameters and the degree of cell transformation. The inhibition of cation transport and ouabain binding to chick embryo fibroblasts infected by RAV but not transformed was markedly less than the inhibition found in the oncogenically transformed cells. These results suggest that alterations in cation transport are intimately related to oncogenic cell transformation. |
