Popis: (1) The characteristic properties of a pyrogenic substance produced in the reaction of delayed hypersensitive cell extracts with antigenin vitro (the so-called hypersensitivity pyrogen) were described and compared with the properties of the leucocytar pyrogen and bacterial endotoxin. (2) The hypersensitivity pyrogen differs from both of them by the typical course of the temperature curve and probably also by the mechanism of fever production; in contrast with the above two substances, the effect of the hypersensitivity pyrogen is inhibited by the immobilization of rabbits. The hypersensitivity pyrogen further differs from endotoxin by its effect in endotoxin-tolerant rabbits. (3) Thein vitro prepared hypersensitivity pyrogen can be precipitated by ammonium sulphate in 20 and 40 per cent saturation. Serum endogenous pyrogen formed on antigen injection to delayed hypersensitive animals is precipitated under the same conditions and elicits a similar type of fever. (4) It is evident from the analysis of the conditions of formation, mode of action and properties, that the hypersensitivity pyrogen is not identical with the leucocytar pyrogen, but that it is a special characteristic substance produced in reactions of delayed type hypersensitivity. (5) These results are discussed from the point of view of the mechanism of delayed type hypersensitivity and of the effect of bacterial endotoxins.
