A method has been developed for quantitatively estimating the contribution of pools of thymine derivatives to the DNA synthesized by bacteria grown in the presence of the analog 5-bromouracil. A 3′-nucleotide digest of the DNA synthesized during a period of labeling with 32P and 5BU is resolved by thin-layer chromatography to determine the 5BU T molar ratio in the newly synthesized DNA. The method is applicable to the quantitative estimation of turnover and repair in the DNA of growing cells. |
