Popis: | The potency of several compounds in inhibiting the autmatically induced by epinephrine in the rat ventricular strip has been evaluated. A correlation with high statistical significancee has been found between pharmacological activity and molecular volume (Mullins' hypothesis) or solubility (Ferguson's rule). These observations have been taken as evidence that the physical properties of the compounds studied are important in determining their potency as inhibitors of ventricular automaticity. Acetylcholine showed high specificity in inhibiting the epinephrine-induced authorhythmicity. It is concluded that this cardiac action of epinephrine may be inhibited not only by antagonists of beta-receptors, but also by a specific action of acetylcholine and by the nonspecific action of various compounds, for which the physical properties of the molecules assume a prominent importance. |
