Interdependence of pH and electrolyte concentration optima for acid deoxyribonuclease activity

Popis: The pH and electrolyte requirements of mouse spleen acid DNase were studied by the methyl green and acid-soluble degradation product methods. The two determinants were found to be interdependent, so that with decrease in pH the concentration of electrolyte required for maximal activation increased. Analysis of this interdependence led to description of the combination of pH and electrolyte concentration which provides maximal possible activation. Different cations and anions differ in their activation effects; in general, bivalent cations are more effective than univalent, and univalent anions are more effective than bivalent. Within each group, systematic differences were observed.
