Popis: | The thermal stability of ribosomes isolated from Escherichia coli grown in minimal medium at different temperatures has been determined by measuring the hyperchromicity at 260 nm while gradually increasing the temperature of the ribosomes which were suspended in standard buffer containing 0.01 M Mg++ions. The melting profiles are dependent upon the sample position in the spectrophotometer since the ribosome suspensions become turbid as the temperature is increased. The measurements did not show any difference between the Tm values obtained for ribosomes isolated from cells grown at 15° C and 37° C, respectively, but at temperatures above 71° C the denaturation profiles of 70S ribosomes from E.coli grown at 15° C and 37° C are different. |
