Contributions of peroxidase immunocytochemistry in the ultrastructural detection of membrane bound antigens

Popis: Over the last 20 years specific membrane polypeptides, such as receptors, enzymes, and organelle-specific antigens, have been visualized in the electron microscope with 'pre-embedding' or 'postembedding' methods of tissue processing (Anderson et al., 1978; Louvard et al., 1982; Tokuyasu, 1986; Brown and Farquhar, 1989; Gonatas, J., et al., 1989; Roth, 1989; Chen et al., 1989). With 'pre-embedding' methods, tissue sections or cells are fixed and 'permeabilized' with a suitable detergent prior to the application of the labeling probe; the labeling probe is then visualized in the light and electron microscopes with horseradish peroxidase (HRP), which may be coupled directly to the labeling probe, or to a second ligand (Guesdon et al., 1979; Brown and Farquhar, 1989). The detection of HRP in the light and electron microscopes is then accomplished with the classic diaminobenzidine (DAB) reaction of Graham and Karnovsky (1966).
