Popis: | Extracellular glucans produced by various strains of oral streptococci cultured on sucrose were separated into two insoluble glucan fractions (IG-1; IG-2) and into a soluble glucan preparation (SG) based on their solubility in water. They were examined by infrared spectrophotometry, acid hydrolysis, acetolysis, periodate-oxidation and Smith degradation. The results suggested that most of the glucans contain α-1:6 and α-1:3 glucopyranosyl linkages, except for some which contained a small quantity of α-1:4 glucopyranosyl linkages. The amounts of α-1:6 linkages averaged 56 per cent for SG, 49 per cent for IG-2 and 38 per cent for IG-1. The percentages of α-1:3 linkages were 37 per cent for SG, 46 per cent for IG-2 and 57 per cent for IG-1. The presence of linear portions containing either α-1:6 or α-1:3 linkages was demonstrated by paper Chromatographic identification of the isomaltoand nigero-oligosaccharides in partial degradation products of the glucans. The capacity to produce each glucan was compared among strains. All strains, except for Streptococcus mitis ATCC 12396, synthesized glucan from sucrose, and all strains of Streptococcus mutans produced the three types of glucan. The main glucan produced by mucoid type strains of Strep, mutans was IG-1; rough type strains of Strep. mutans found mainly SG. Streptococcus sanguis ATCC 10557 produced SG, but it entirely lacked IG-1 and IG-2-producing capacity. The possible relationship between the percentage of α-1:3 glucopyranosyl linkages in the glucans and their alkali-solubility was examined. The results strongly suggest that the solubility of the glucans depend on factors such as their structural features as well as their content of α-1:3 glucopyranosyl linkages. |
