Popis: | Medroxyprogesterone acetate, chlormadinone acetate, AY-11,497 and clogestone acetate (AY-11,440) reduced adrenal and ovarian weights, serum and adrenal corticosterone in the rat. Adrenal homogenates from rats treated with medroxyprogesterone acetate showed impaired capacity to convert progesterone (IIβ- and/or 21 -hydroxylation) and 11β-hydroxyprogesterone (21-hydroxylation) to corticosterone. In vitro, medroxyprogesterone acetate and clogestone acetate (1 × 10-4M) did not affect the conversion of progesterone to corticosterone. Estradiol-17β treatment enhanced the conversion of progesterone to corticosterone by adrenal homogenates from most progestogen treated rats, and the conversion of 11β-hydroxyprogesterone to corticosterone by adrenal homogenates from rats treated with medroxyprogesterone acetate.
