Popis: | In a recent paper, Garlic [l] described the application of a new iron-chelating ligand, chromazurol B (CAB), in a manual method for the direct determination of serum iron and total iron-binding capacity. In this method, Fe(III)/Fe(II) reacts with CAB and cetyl trimethylammonium bromide (CTMA) to give a highly coloured ternary complex with a molar extinction coefficient of 1.68 X 10 5 1. mol ’ . cm’ at 630 nm. As this method, in our experience, gave a low percentage of recovery, we partially modified the original reagent composition and performed the reaction at 37°C instead of at room temperature. We also eliminated the preparation of CAB ammonium salt, by solubilizing the acid CAB directly in the reagent in the presence of ethanol, thereby obtaining a single, stable reagent. |
