Popis: | Hydroxyproline was found in both young and old rat’s cells obtained from kidney cortex by means of trypsinization. Its amount was much lower than the amount of hydrozyproline in the tissue remaining after the separation of the cells. In the remaining tissue, the amount of hydroxyproline was higher in the tissue from old donors. In the trypsinized cells, the same difference was apparent when the values were calculated per 100 μg nitrogen, not per 100 mg dry wt. Further treatment of the trypsinized cells with collagenase resulted in a decrease of hydroxyproline in both samples of cell to negligible values. In spite of this, the cells from old donors showed evidence of damage in culture. Only isolated elements of abnormal shape appeared under conditions allowing formation of many colonies of epithelial cells in cultures from young donors. |
