Popis: | A detailed study was made of the structural changes in the genital organs of rabbits and dogs given KABI 1774 (cyclotetrasiloxane). The compound was given orally in doses of 2 mg/kg body weight daily for periods of from 2 to 30 days. For controls 15 rabbits were given oil only and 8 rabbits were given estradiol benzoate 7.5 mcg/kg sc for periods of 5-20 days. A study was also made of the epididymides of 4 rabbits at periods of 2-9 days after castration. 8 dogs were given 10 mg or 250 mg/kg of the cyclotetrasiloxane for 40 days. Tissues obtained at autopsy were prepared and studied by both light and electron microscopy. After 2 days of therapy in the rabbits some vacuolated cells with necrotic nuclei were seen in the epididymal epithelium especially in the middle segment. After 15 days of treatment all genital tissues showed some alterations with variations between rabbits. 4 rabbits showed disturbance of spermatogenesis and all had altered Leydid cells. All regions of the epididymal duct showed degenerating and dead epithelial cells. Spermatozoa no longer showed normal maturation changes. After 20 days of therapy spermatozoa in the cauda epididymis were dead and the seminiferous tubules showed arrested spermatogenesis. In the dogs the effect varied from arrested spermatogenesis to complete degeneration of germ cells and atrophy of intersitital cells. The dogs prostate glands showed epithelial atrophy. The epithelium of the ureter and urinary bladder were unchanged. The kidneys and adrenals remained normal but the liver showed some changes after 20 days of treatment. Variations between individual dogs were more marked than differences between those given different doses. Epididymal atrophy was less marked than in rabbits. The epididymides of rabbits after castration showed similar epithelial changes as those caused by the cyclotetrasiloxane treatment for 10-25 days. In the 8 rabbits given estradiol benzoate after 20 days some epithelial changes were observed. Several reproductions illustrate microscopic findings. The study confirms the sensitivity of the rabbit testis and epididymis to this drug. The accessory genital glands in rabbits were less sensitive but the dogs prostate showed marked sensitivity. |
