Popis: other major group of regulatory proteins for Ras, have been mostly attributed a rather passive role related to termination of Ras-elicited signals. In this review we have attempted to summarise our current understanding of the reactions that lead to the cellular accumulation of Ras-GTP. One major focus will be the discussion of emerging concepts for the action of recently discovered RasGEFs which have expanded (and at the same time possibly complicated) the overall picture of the control of Ras activation. In the second part of the review we discuss an alternative to GEF activation, namely GAP inhibition, as a means to induce the accumulation of Ras-GTP; this part presents a summary of experimental data that point to an active role of GAP proteins in the agonist-induced formation of Ras-GTP.
