Popis: | It is confirmed that pyrazole prevents the sleep-inducing effect of butane-1, 4-diol in mice. In addition, it is shown that pyrazole potentiates 4-hydroxybutyrate-induced sleep but has no effect on succinic semialdehyde-induced sleep. Pyrazole itself has a slight sedative action at doses above 450 mg/Kg. Mouse liver extract and liver alcohol dehydrogenase readily oxidise butane-1, 4-diol (K M 1.6 × 10 −3 M). This oxidation is competitively inhibited by pyrazole (K I 5.4 × 10 −7 M). It is proposed that this inhibition provides the basis for the interaction between pyrazole and butane-1, 4-diol. |
