Supplementary Figure 1: CBS9106 reduces CRM1 protein levels through neddylation -mediated degradation in MM.1S cells. (A) MM.1S cells are treated with CBS9106 (50 nM) and/or MLN4924 (100 nM) for indicated period (0-8 hours). (B) Cells are treated with increasing concentrations of MLN4924 (0, 10, 20, 50, or 100 nM) and/or CBS9106 (50 nM) for 24 hours. (A, B) Protein levels of CRM1, CyclinB1, CulNEDD8, p53, PARP, p27, CDT1, and beta-actin are determined by western blot analysis. The measured band density was normalized relative to that of the control sample, with the control value set to 1. MM.1S cells was kindly provided by Dr. H. Yasui (University of tokyo, Tokyo, Japan).
