Association of adenovirus early proteins with a nuclear fraction that synthesizes DNA in vitro

Popis: We describe a soluble subnuclear fraction isolated from adenovirus type 5-infected HeLa cells which when incubated in vitro synthesizes adenovirus DNA. This synthesis is inhibited by immunoglobulins directed against virus-specified proteins synthesized early in infection, but not by immunoglobulins directed against the viral coded tumor antigen (T antigen). Analysis of the fraction on sucrose gradients demonstrates that it contains two distinct nucleoprotein complexes. One of these complexes sediments at about 100 S and contains replicating viral DNA molecules. The other, which sediments at about 40 S, contains mature double-stranded DNA of genome length. After a pulse-label with [ 35 Simethionine, the virus-coded 72 K single-stranded DNA binding protein (DBP) is found associated with the 100 S peak but not with the 40 S peak. Following a chase period, however, the DBP also appears in the 40 S peak. Labelling with 32 PO 4 also shows that the phosphorylated form of the 72 K DBP is found initially only in the 100 S peak. These results suggest that newly made and phosphorylated DBP associates preferentially with replicating viral DNA.
