Bile acids and lipid metabolism. IV. Influence of bile acids on biliary and liver organelle phospholipids and cholesterol

Popis: The excretion of C 14-bile acids, P 32-phospholipids, and C 14-cholesterol (mass and radioactivity) in the bile of the isolated perfused rat liver was determined with and without the infusion of sodium taurocholate. The liver cholesterol and phospholipids were labeled prior to the infusion of taurocholate. There was very little excretion of bile acids and lipids in the bile without the taurocholate infusion. Taurocholate infusion (for 1 or 2 hours) markedly increased the biliary excretion of P 32-phospholipids and C 14-cholesterol. Examination of the P 32-phospholipid fraction indicated that 94-97% of the P 32-activity in the bile was in the phosphatidyl choline fraction. The specific activity of the bile phosphatidyl choline exceeded the specific activity of that fraction in the liver mitochondria and microsomes. There was also a marked heterogeneity in the labeling of the liver organelle and plasma phospholipids. The results suggest that bile acids in the enterohepatic circulation regulate the excretion of biliary lipids. This may be related to the formation of a specific macromolecular complex in the liver and its subsequent excretion in the bile. (Author)
