The amino acid sequence of cerato-ulmin (Stevenson et al., 1979), a toxin produced by Ophiostoma (Ceratocystis) ulmi (Buisman) Nannf., the causal agent of Dutch elm disease, has been completely determined. Cerato-ulmin consists of two regions (residues 1–38 and 39–75), and each contains two disulfide bonds. The molecular weight calculated from the amino acid sequence is 7618.7 and the molecular weight determined by Ion-Spray Mass Spectrometetry using a Sciex API III mass spectrometer is reported to be 7618.4 (Feng et al. 1991). The infrared and Raman spectra show that the protein contains about 15% alpha-helix and about 50% beta-sheet. The Raman spectra also revealed no evidence for an S-H stretching vibration near 2580 cm-1 but an intense S-S stretching mode is apparent at 514 cm-1. Thus all eight cysteine residues are linked in disulfide bonds. |
