Genetic recombination in Escherichia coli B III. The influence of experimental conditions on the transfer of unselected markers

Popis: a. The transfer-percentage of some unselected markers can be influenced by experimental conditions such as the nature of the culture medium on which the F− parent is grown. This effect varies for different loci. b. Introduction of other markers may abolish the effect mentioned undera. Cultural conditions can therefore influence the apparent degree of linkage. c. Addition of methionine to recombination plates can materially alter the percentage of transfer of unselected markers. This effect should be taken into consideration in studying linkage to the methionine locus. For such studies the usual procedure of comparing prototrophs (m+) from recombinations on minimal medium with recombinants (m−) on methionine supplemented plates is dangerous. To arrive at more reliable linkage data the m+ class should also be derived from supplemented plates.
