Popis: | Heterogeneity of carbohydrate lectin binding sites in carcinoembryonic antigen (CEA)-related preparations was determined by a Farr type radioimmunoassay using the lectins concanavalin A and wheat germ agglutinin. The technique was shown to involve a specific interaction of lectin and carbohydrates as only the relevant monosaccharides were capable of inhibiting CEA precipitation by lectin. Distinct differences in the titration curves of lectin binding were shown for several CEA-related preparations including colon cancer-derived CEA, ovarian cancer-derived CEA and CCA-III. Our results indicate that these lectins can be used as additional probes for the characterization of tumor-associated glycoproteins such as CEA preparation derived from various sources. |
