Metabolism of Butyl 4-Hydroxy-3,5-diiodobenzoate BHDB), a Thyroxine Analogue

Popis: The metabolism of butyl 4-hydroxy- 3,5-diiodobenzoate (BHDB) labeled with 13II in the 3,5 position and that of thyroxine (T4) labeled with 131I in the 3',5' position were compared in isolated liver (rat) perfusions and in rats with biliary fistulas. Disappearance of radioactivity from the blood was more rapid, biliary excretion of radioactive compounds was greater, and accumulation of 131I in the liver was less after administration of BHDB than after administration of T4. Biliary and urinary metabolites of BHDB in whole rats were similar to biliary metabolites from perfused livers although the metabolites did not accumulate in blood perfusing the isolated liver. Deiodination and glucuronide or sulfate conjugation were not found to be pathways of BHDB metabolism. An acid-hydrolyzable conjugate of BHDB was found in bile and urine but was not identified. In spite of their similarities in molecular structure, BHDB and T4 are metabolized by different pathways. (Endocrinology 83: 239, 1968)
