Popis: | Measurement of endogenous thyroid stimulating hormone (TSH) in monkey plasma by radioimmunoassay technique is described. This method exploits the ability of endogenous monkey TSH to inhibit competitively the binding of 125I-labeled human TSH to antibodies to human TSH. The sensitivity of this procedure is sufficient to detect TSH in 0.4 ml of serum of fasting monkeys. In terms of the Human Thyrotropin Research Standard A, normal plasma TSH level of the fasting adult male monkey was found to be 6.8±2.3 μU/ml. Three weeks post thyroidectomy the TSH level rose to 168 μU/ml. After a single injection of 100 ng thyroxine in the thyroidectomized monkey the TSH value fell from 162 to 54 μU/ml in 1 day and to 12 μU/ml by 3 days. The labeled human TSH was suitable for assay after addition of an anion exchange resin to remove iodide without further purification. (Endocrinology 86: 769, 1970) |
