Popis: | IN Escherichia coli1 and Bacillus subtilis2, the most recently synthesized DNA was found to be attached to the cell membrane. When human amnion tissue culture cells were briefly exposed to3H-thymidine, electron microscope autoradio- J graphy of the cells showed that the radioactivity was restricted to the nuclear envelope and the periphery of the nucleolus. This suggested that DNA replication was initiated at the nuclear membrane3. We have analysed DNA synthesis in nuclear envelopes isolated from rats shortly after administration of3H-thymidine and have shown that nascent DNA, as indicated by DNA of highest specific activity, was associated with a membrane fraction derived from the inner layer of the nuclear envelope. |
