Popis: | Dimethylsulphate treatment of 1-β-D-arabinofuranosylcytosine (ara-C) in aqueous alkaline medium, followed by column chromatography, has led to isolation of all possible O'-methylated derivatives of ara-C. The three mono-methylated derivatives were obtained in crystalline form and identified on the basis of NMR spectra and other criteria. The 3′,5′-di-O-methyl derivative was distinguished from the 2′,5′- and 2′,3′- on the basis of its susceptibility to acid-catalyzed deamination. The potential antimetabolic properties of these new ara-C analogues are discussed. |
