Popis: | A fragment of Escherichia coli DNA carrying a nonsense suppressor transfer RNA gene was joined in vitro to a fragment of simian virus 40 DNA containing the origin of viral DNA replication and early gene region. This recombinant molecule was replicated and encapsidated into virions in monkey cells coinfected with suitable helper simian virus 40 DNA. Structural studies showed that the E. coli DNA suffered no observable sequence alterations. Monkey cells infected with the recombinant virus transcribe the bacterial DNA, but fail to synthesize detectable amounts of mature suppressor tRNA. |
