Popis: Metal-catalyzed, bidentate directing group-assisted, C–H activation and functionalization have expanded exponentially in the past few years and provides numerous opportunities in the synthesis of a range of substrates in a straightforward and step-economic manner. Various bidentate auxiliaries have been introduced for the functionalization of C(sp2)–H and C(sp3)–H bonds to C–C and C–X bonds and applied to the synthesis of complex, bioactive natural products in an efficient manner. This approach reduces the synthetic difficulty of establishing complex architectures by by-passing the pre-functionalization of starting materials. Herein is presented various bidentate directing groups and their use in the metal-catalyzed, C–H functionalization in the total synthesis of complex natural products. Various bioactive natural products, including cyclobutane molecules, complex peptides, lignan molecules, gut metabolites and β-lactone derivatives were successfully synthesized in a short route. Essentially, the core fragments or the key reactions of these natural products were successfully accomplished using bidentate auxiliaries.
