Identification and characterization of two piwi genes and their expression in response to E2 (17β-estradiol) in Dabry’s sturgeon Acipenser dabryanus

Popis: | Piwi proteins are essential for germline stem cell maintenance, gametogenesis and gonadal development in diverse organisms. In this study, two piwi homologues were identified and characterized in Dabry’s sturgeon Acipenser dabryanus, designated Adpiwil1 and Adpiwil2. Multiple sequence alignments showed that both AdPiwil1 and AdPiwil2 shared highly homologous PAZ and PIWI domains, the typical regions of Piwi orthologs. Phylogenetic analysis revealed that AdPiwil1 and AdPiwil2 were conservatively grouped with teleosts. We found that the Adpiwil1 transcripts were abundantly expressed in gonadal tissues and rarely present in brain tissues, whereas the Adpiwil2 transcripts were found to be specifically expressed in gonads by tissue distribution analysis. Fluorescence in situ hybridization also revealed that both Adpiwil1 and Adpiwil2 mRNAs were restricted exclusively to germ cells of both sexes. In addition, E2 (17β-estradiol) intraperitoneal injection was performed on female Dabry’s sturgeons, and its effects on Adpiwil1 and Adpiwil2 mRNA levels were evaluated by relative real-time PCR. The results showed that the transcription levels of Adpiwil1 and Adpiwil2 were significantly decreased in ovaries after E2 injection. Our findings will contribute to an understanding of the piwi function and provide a basis for further studies. |
