Popis: | The effects of two anabolic steroids, stanozolol and methenolone, on gene expression and protein secretion of apolipoprotein A-I and apolipoprotein B-100 were analyzed in the human hepatoma cell line HepG2. Concentrations of mRNA for apolipoproteins A-I and B-100 were determined by quantitative slot blot hybridization. The amount of apolipoproteins secreted into the media were measured by zone immuno electrophoresis or enzyme linked immunosorbent assay. At concentrations of 10–9 M to 10–7 M no effect on mRNA concentration or apolipoprotein secretion could be detected. At higher concentrations of the anabolic steroids non-specific toxic effects on the HepG2 cells were observed. It is concluded that the known effects of anabolic steroids on lipoprotein metabolism in vivo, i.e. lowering of high density lipoprotein cholesterol and apolipoprotein A-I and increase of low density lipoprotein cholesterol and apolipoprotein B-100, is not initiated by significant changes in synthesis and secretion of the respective apolipoproteins. |
