Popis: Rats and mice have become the models of choice for the study of a wide range of metabolic bone diseases. One key outcome measure used to evaluate skeletal physiology in these models and to assess response to treatment is histomorphometry. This chapter describes the essentials of proper specimen preservation and preparation as well as static and dynamic histomorphometric methods used to evaluate modeling and remodeling in rodent models. Approaches to histological evaluation of skeletal microdamage and its repair are discussed. The chapter reviews the phases of fracture healing and discusses histological approaches for assessing the remodeling and maturation of fracture callus. These methods are then discussed with reference to common rodent models of bone loss and pharmaceutical treatments, with an evaluation of the strengths and weaknesses of rat and mouse models for these applications.
