Popis: Summary The replication of herpes simplex virus in human leukocyte cultures has been demonstrated in several ways. A consistent decrease in viral titer was observed over the first few days, usually followed by an increase in titer. Virus was demonstrable in such cultures for as long as 26 days. A viral inoculum of at least 103 TCD50 per 0.2 ml of culture was found necessary to initiate infection. Viral replication did not occur in the leukocyte cultures in the absence of phytohemagglutinin. However, viral multiplication was not affected by certain variations in technique of leukocyte culture preparation or incubation, or by age of the culture at time of viral inoculation. The kinetics of herpes simplex virus replication in human leukocyte cultures are discussed.
