Crystal structure, DNA binding, cytotoxicity and anticancer ability of Zn(II) complex constructed by 2-(1,2,4)triazol-1-yl-isonicotinic acid

Popis: | A novel Zn(II) complex was designed and synthesized under hydrothermal condition based on the ligand 2-(1,2,4)triazol-1-yl-isonicotinic acid. The crystal data were collected by X-ray single crystal diffractometer, and the purity of the complex was verified using elemental analysis and powder X-ray diffractometer. UV–Vis spectroscopy analyzed the binding mode of the complex and CT-DNA and calculated the binding constant Kb value of 3.36 × 103 M−1. Fluorescence spectroscopy further studied the interaction mode of the complex with CT-DNA, and the quenching constant Ksv value was 0.06. Gel electrophoresis was used to study the ability of the complex to cut pBR322 plasmid DNA. Flow cytometry detected the apoptosis ratio of the complex to HeLa cells, and the total apoptosis rate reached 71.4%. The fluorescent inverted microscope was used to observe the morphological changes of the cells by double staining, and detect the apoptotic effect on HeLa cells. |
