The Observation of Nematolysosome in Neuron of Anterior Horn in Guinea Pig Spinal Cord by Enzyme Cytochemistry and Its Quantitative Energy Dispersive X-Ray Analysis

Popis: In this study, the presence of nematolysosome (NLY) with trimetaphosphatase (TMPase) activity and acid phosphatase (ACPase) activity in the neuron of anterior horn in guinea pig spinal cord was certified by means of enzyme cytochemistry. NLYs with TMPase and ACPase activity were observed in soma, process and synapse, where NLYs attach to mitochondria tightly. The observations we made revealed that NLY could transport from soma to nerve terminal, which chiefly depended on mitochondria to provide energy for the movement of NLYs. Our findings implied that NLYs may be related to the decomposition of neurotransmitters, the disposition of metabolites and the TMPase and ACPase transport in neuron. Meanwhile, the quantification of the TMPase and ACPase activity of NLYs by energy dispersive X-ray analysis (EDAX) which indicated that the activity of TMPase was higher than that of ACPase, completely complied with the observation of cytochemistry that the number of NLYs with TMPase activity was much more than that with ACPase activity. Our results cytochemically and quantitatively showed that as a lysosomal maker enzyme, TMPase is more sensitively demonstrated than ACPase in the neuron of anterior horn in guinea pig spinal cord.
