We have investigated the in vitro effect of adenovirus (AV) infection of epithelial cells on intracellular distribution of SSB antigen. Hep-2 cells were infected with AV-II and the SSB antigen localization was studied at various time intervals using a monospecific anti-SSB antibody and an indirect immunofluorescence (IIFL) method. Pooled normal human IgG was used as control. After 3 hrs the antigen concentration was decreased in nuclei and increased in cytoplasm. After 24 hrs, the SSB-antigen appeared extracellularly in form of small granules. These granules formed complement fixing immune complexes in the presence of anti-SSB antibody and fresh human serum. This was demonstrated by a fluorescent anti-C3 reagent. Control immunoglobulin at identical conditions of IIFL gave a negative reaction. Highly purified SSB antigen-antibody complexes were prepared and were shown to activate substantial amounts of complement. |
