The effects of dilution rate (D) on I.13.17. hybridoma growth, metabolism, and antibody production in semi-continuous culture were evaluated. Steady-state data were obtained at six D between 0.008 and 0,055 h-1. Viable cell concentration was maximal at intermediate D (0.03 h-1). The specific growth rate was significantly affected by the D, particularly at lower D. Monoclonal antibody (MAb) concentration and specific MAb production rate were maximum at low D, during periods of slower growth. The cellular metabolic activity was observed to increase linearly with increasing D. |
