Popis: | Since little information is available explaining the low conversion frequency of transient to stable transformation events, we analyzed particle location and cell viability after particle bombardment. Cotyledonary explants and immature zygotic embryos of sunflower were used as targets. In order to optimize the biological interaction physical parameters and the target tissues, two different particle DNA acceleralion systems as well as explants of various developmental stages were used. Sunflower (Helianthus annuus L.) has proven to be recalcitrant in tissue culture, especially con-cerning genetic transformation. Until now, four publications describe successful stable integration of foreign genes in regenerated R0 plants (1–4), but only two of them demonstrated the transmission of introduced genes to the next generation (3–4).
