The virions of adenovirus type 5 (Ad5) DNA contain a linear double-stranded DNA molecule with a length of 11 ym corresponding to a molecular weight of 23 million daltons (Eb, Kesteren, and Bruggen, 1969; Green et al., 1967). Pearson and Hanawalt (1971) have shown that by labeling adenovirus-infected HeLa cells with 3H-thymidine for 5 minutes, the major fraction of the label entered into a fast-sedimenting “replication complex.” The label could be chased from this complex into mature adenovirus DNA. The DNA isolated from these complexes showed an increased buoyant density in CsCl as compared to mature adenovirus DNA. This suggested the presence of single-stranded DNA or RNA in the replicative forms of adenovirus DNA. |
