Measurement of DDT and DDE from serum, fat, and stool specimens

Popis: Dichlorodiphenyltrichloroethylene (p,p'‐DDT) and metabolites (DDE and DDD) are identified and measured in human tissues. Liquid‐liquid and solid‐phase extraction procedures from serum, fat, and stool samples are presented and the resultant extracts are quantitated using capillary gas chromatography and electron capture detection. For serum, the linear range is from 1 to 500 ppb (ng/mL). Using a four mL specimen and liquid‐liquid extraction the expected accuracy is within ±15% for each analyte, with an extraction efficiency ranging from 92 to 101%. For stool samples, the linear range is 20–2000 ppb for DDE and 20–400 ppb for DDT. Using 4 gram of stool specimen the accuracy is within ±11% for each analyte. Extraction efficiency ranged from 92–111%. Using adipose tissue and extracting fat by solid‐phase C18 columns the extraction efficiency using 100 mg fat sample ranged from 70–95%. If required, DDT and metabolites are confirmed by electron ionization gas chromatography‐mass spectrometry.
