When an immobilized enzyme is used to catalyze a reaction in which oxygen is a substrate, there are three principle mass transfer steps which may cause problems (Table 1). It is possible to prepare immobilized oxidases with activities of 0.5 μmol/mg/min or greater, which are higher than the oxygen demands of rapidly growing microorganisms. Thus the same problems of transfer of oxygen from the gas to the liquid phase can arise in immobilized oxidase reactors as occur in aerobic fermentations. The rate of oxygen transfer depends on the gas flow, amount of agitation and the oxygen partial pressure in the gas phase. The agitation may cause attrition of the immobilized enzyme particles. The effect of shear on immobilized enzyme activity and particle size has already been described (1). |
