Members of the tumor necrosis factor receptor superfamily (TNFRSF) play essential roles in tissue development and homeostasis. Through unique receptor–ligand interaction, TNFRSF members become activated and recruit intracellular adaptor proteins to initiate diverse signaling pathways mediating cell survival, proliferation, differentiation, and apoptosis. Deregulation of TNFR pathways is associated with inflammatory and autoimmune disorders as well as cancer. Therapeutic strategies of targeting TNFRSF members and their ligands have been developed. |
