A WHO cooperative trial in hypercholesterolemic patients treated with clofibrate (Cf) shows a higher index of mortality in treated patients (Lancet, 378, 1980). A previous hyperlipidic diet (HL) (butter, cholesterol, cholic acid, thiouracil) protects the rats against the atherosclerotic lesions induced by vit D2; contrarily, the administration of HL following to vit D2 impairs the lesions, reaching an optimal experimental model of atherosclerosis (AT) (Villaverde et al. Proc. X I.G. Angiology, 285, 1976). We have studied the implication of the biochemical changes induced by HL, and the influence of Cf on the experimental model AT in rat. The HL, that did not produce vascular injury during the period of experiment, induced an increase in cholesterol, from 76 to 717 mg/dl, changing the total-cho/HDL-cho ratio (cho-R) from 1.1 to 8.7.
