Popis: | It was found that in addition to its hypoglycemic action 1-propyl-3-p-chlorobenzene- sulfonylurea has an antidiuretic effect on patients with diabetes insipidus as well as on normal subjects. This effect was studied by the intravenous injections of chlorpropamide, comparing the ensuing diuresis with that obtained in basal conditions and after the intravenous administration of a solution of 2.5% NaCl, vasopressin and diuretics of the thiazide type. The tubular management of water studied by measuring osmolal clearance and clearance of “free” water revealed that chlorpropamide decreases diuresis, increases the osmolality of urine and decreases the clearance of “free” water which may become negative, without affecting the glomerular filtration rate or osmolal clearance. These results lead to the conclusion that chlorpropamide has an antidiuretic action similar to that of vasopressin. The oral administration of chlorpropamide decreases diuresis in patients with diabetes insipidus, rendering unnecessa... |
