Popis: We have developed novel injection schemes to define extremely small sample plugs, thereby allowing ultra-high speed DNA separations. One method uses a second intersection to prepinch the sample, and in a second method, called reverse injection, the sample loading direction is briefly reversed before injection. As an example of the improvement resulting from the use of very small sample plugs, we have demonstrated separations of ssDNA oligos with single-base resolution in less than 4 seconds. We believe that these are the fastest DNA separations on a microchip demonstrated to date.
