Popis: Publisher Summary pBluescriptll phagemid vectors are multifunctional cloning vectors designed to simplify and expedite gene cloning and analysis. The vectors contain polylinkers that are designed to optimize a number of cloning procedures including directional cloning, gene mapping, unidirectional deletions of predictable length, T3 and T7 polymerase-mediated transcription, blue/white color selection for clones with insert, double-stranded DNA sequencing, and β-galactosidase fusion protein expression. pBluescript vectors contain an ampicillin resistance gene. pBluescript vectors replicate at a high copy number owing to the lack of the pBR322 rom gene. The versatility of pBluescript vectors alleviates the need for successive subcloning. With 21 restriction sites, in two orientations, most cloned genes can be inserted directionally into the pBluescript polylinkers. Polymerase chain reaction-amplified fragments from RNA, cloned DNA, or genomic DNA can be inserted directly into pBluescript vectors, bypassing the need to construct large libraries. The restriction enzyme sites in the pBluescript polylinkers are arranged in a manner that allows unidirectional deletions of inserts with exonuclease III.
