On-line measurement of OUR using off-gas analysis equipment has been used to implement monitoring and advanced control schemes in several microbial fermentation systems. The development of similar applications for cell culture systems has been limited by the difficulties of measuring OUR directly using a gas mass balance in bench scale fermentors. In this study, we developed on-line measurement of OUR based on a liquid mass balance for headspace aerated cultures. A Kalman filter was used to extract the oxygen uptake rate from the measured liquid oxygen concentration and the controlled oxygen concentration in the gas inlet. A dynamic matrix control strategy was used to ensure smooth oxygen control, thus improving the accuracy of the estimator. |
