Popis: Publisher Summary This chapter discusses the brain microvessel butyrylcholine estarase in ischemia and postischemia. It discusses a study to investigate the presence of large amount of butyrylcholine esterase in brain capillaries. The enzyme was localized in the cytoplasm of endothelial cells in blood-brain barrier protected areas of the CNS, while in neuropil, the product of histochemical reaction was discrete. Entorhinal cortex appeared to be unique region, where abundant reaction product was found in both pericarya and processes of pyramidal cells. Transient cerebral ischemia was produced in Mongolian gerbils and the changes in localization and activity of BuChE were followed in reflow in freshly frozen brain sections and isolated brain capillaries using the histochemical and biochemical procedures, respectively. Histochemically, in ischemia and in immediate reflow, the discrete reduction of BuChE product was found in microvessels, contrary to the significant reduction of the enzyme activity obtained by neurochemical procedure. At 7 days reflow period, both histochemical and biochemical methods revealed the significant increase of BuChE activity in the microvasculature of cerebral cortex indicative for newly developed brain microvessels.
