GENETIC DIVERSITY OF OLIVES GROWN ALONG THE COAST OF MONTENEGRO

Popis: | We studied the genetic diversity of autochothonous and introduced olive cultivars in Montenegro using isoenzymes and RAPD markers. The isoenzyme analysis was undertaken on 39 accessions of what was thought to be the cultivar 'Zutica', another 22 accessions of autochothonous cultivars and 19 foreign cultivars. A method for protein extraction from inner bark was developed and good enzyme activity obtained for all 14 systems analysed: ADH, CAD (NAD and NADP), ENP, EST, GDH, GPI, GOT, HEX, LAP, MNR, PGM, PRX and SKD. In all, 31 putative loci were proposed. RAPD analysis was carried out on a subset of samples using 13 primers and also revealed a high polymorphism. Isoenzyme and RAPD data were used for cluster analysis (UPGMA) to assess the genetic similarity between individuals analysed. The work demonstrated that 'Zutica' is a mixture of genetically distinct variants and that there is no apparent clustering of olive cultivars according to their geographic origins. |
