A study was carried out to investigate a possible involvement of dicarboxylate transport in the accumulation of PHA by R. meliloti. Mutants derived from the wild-type CM2 were used, which are affected in dicarboxylate transport, namely CM12 (dct B−) and CM2 pCU28, the later having an enhanced transport system for malate, fumarate and succinate. Experiments with shaken cultures and 2 lt-fermentor would suggest that mutants with enhanced transport of dicarboxylic acids accumulate higher amounts of PHA, compared to their parental wild-type strain. |
