It consists of 4 subunits which are homologs of two catalytic subunits of bacterial RNAP, and 5th subunit which have not homologs with a known function. The role of the fifth subunit in the nvRNKP complex is investigated at the laboratory. It was shown that the fifth is necessary for the assembly of active nvPNKP and takes part in interaction with DNA. The limited proteolysis method was used to investigate DNA-protein complex-es of nvPNAP with different types of DNA-matrices. Binding with DNA reduce the formation of a protealized fragment of the fifth subunit corresponding to 1-382 aa. In order to study what else is affected by the C-terminal of the fifth subunit, experiments were carried out to emit the complex formed from it and the 4x subunit polymerase in vivo. It was determined that the C-end of the fifth subunit is necessary during interac-tion with DNA and especially during interacting with "long" DNA, and it is also im-portant for the assembly of enzyme in vivo. |
